At a film history museum, a tour guide tells visitors the incredible story of James and Henry, two Minions whose adventures helped shape early Hollywood.
Long before serving Gru, a different tribe of Minions searched the world for history's greatest villains. Unfortunately, their enthusiasm usually ended in disaster, as they accidentally killed one master after another. Among them was James, a creative Minion with a passion for painting and filmmaking, who formed a close friendship with Henry and Ed. During one adventure, their evil warlock master is devoured by a monster that Henry accidentally summons from the warlock's spellbook, which Ed keeps after the incident.

While wandering through the desert, the trio witnesses what appears to be a train robbery and eagerly chases the outlaw, hoping to become his henchmen. They soon discover it is actually the filming of a Hollywood western. Although director Max is furious at the interruption, studio executives Frank and Elwood are charmed by the Minions' chaotic antics. Max reluctantly casts them in his films, and the Minions quickly become silent-film superstars.
The arrival of talking pictures brings their careers to an abrupt end. Since no one can understand the Minions' language, their sound films are disastrous, costing the studio a fortune and getting them fired. Refusing to abandon his dream, James decides to make his own monster movie, Minions and Monsters. While the tribe's leader, Dick, dismisses the idea and leads the other Minions away to search for another evil master, Henry and Ed stay behind to help James. Max encourages the project by giving James a film camera. Hoping to find a suitable monster, Ed uses the old spellbook to summon a creature, but instead conjures a tiny monster named Goomi. Excited by James' idea, Goomi offers to help them find bigger monsters for the film.
Elsewhere, Dick and the rest of the tribe begin serving Dort, an advanced alien robot determined to conquer Earth. However, Dort's plans are derailed when he falls in love with a women's rights activist named Debbie and enlists the Minions to help win her affection.

Goomi leads James, Henry, and Ed to a frozen fortress, where he releases two sea monsters, Phillips and Howard, under the guise of casting them in the movie. In truth, he plans to use them to summon Irene, a gigantic orange blob monster capable of destroying the world. Henry overhears the scheme but is captured and prepared as the sacrifice needed for the ritual. As filming begins, Irene emerges and immediately goes on a destructive rampage.
While on a date with Debbie, Dort spots the chaos and initially flees, leaving several Minions to battle Irene themselves. During the fight, Irene swallows Henry, forcing James to dive inside the monster to rescue him. Although the Minions briefly manage to tear Irene apart, she quickly regenerates. Just as all hope seems lost, Dort returns with the rest of the Minions aboard a fleet of spacecraft and destroys Irene. James and Henry then use the spellbook to imprison Goomi, Phillips, and Howard inside it. The Minions are celebrated as heroes.

Because Ed filmed the entire adventure, Minions and Monsters premieres several months later to a standing ovation. The museum tour concludes with a final twist: both the story and the museum tour itself are revealed to be scenes from another movie directed by James and Henry.
During the mid-credits scenes, the warlock's spellbook eventually finds its way into the hands of Gru's Minions, where Kevin, Stuart, Bob, and Agnes begin experimenting with its magic.
